Modeling OxLDL Epigenetic Innate Immune Training of Monocytes in Early Atherosclerotic Plaques

چکیده

Atherosclerotic cardiovascular disease (atherosclerosis) is the leading cause of death worldwide. While atherogenesis generally well understood, sustained inflammation seen in some rapidly developing plaques remains largely unexplained. Recent research epigenetics reports chromatin remodeling with significant pro-atherogenic effects human monocytes exposed to oxLDL. Data collected from isolated vitro suggests that epigenetic innate immune training may explain observed persistent inflammatory state, however, has yet quantify on early plaque dynamics. In present study, I employ a partial differential equation and agent based mathematical model describe key markers system “untrained” or “trained” monocytes. Time dependent solutions suggest oxLDL produces pronounced pro-inflammatory response which counts macrophages foam cells. These results provide further support for targeting epigenome treatment atherosclerosis.

OxLDL-IgG immune complexes induce survival of human monocytes.

OBJECTIVE Immune complexes containing oxidatively modified low-density lipoprotein (oxLDL) particles are deposited in human atherosclerotic lesions during atherogenesis. Here we studied whether OxLDL-IgG immune complexes (OxLDL-IgG ICs) affect survival of human monocytes. METHODS AND RESULTS As demonstrated by light microscopy, and analysis of cell proliferation, caspase-3 activity, and DNA f...

Effect of Helicobacter pylori DNA in human atherosclerotic plaques

Introduction: A number of studies have demonstrated that infectious mico organisms like helicobacter pylori may play a role in the process of atherosclerosis. We, here, aimed to investigate the effect of Helicobacter pylori DNA in atherosclerotic plaques in patients with coronary artery disease. Methods: In a cross-sectional study, 85 patients undergoing coronary artery bypass graft (CAB...

Prevalence of Atherosclerotic Plaques in Autopsy Cases with Noncardiac Death

Background and Objective: Ischemic heart disease (IHD) following atherosclerosis is the most common cause of cardiac deaths world wide. We aimed to investigate the pathologic features of atherosclerosis in non cardiac death cases to have an estimate of atherosclerosis prevalence in Mashad (North east of Iran). Patients and Methods : This descriptive (cross sectional) study was done, during ...

Modeling Innate Immune Response to Early Mycobacterium Infection

In the study of complex patterns in biology, mathematical and computational models are emerging as important tools. In addition to experimental approaches, these modeling tools have recently been applied to address open questions regarding host-pathogen interaction dynamics, including the immune response to mycobacterial infection and tuberculous granuloma formation. We present an approach in w...

Early atherosclerotic plaques show evidence of infection by Chlamydia pneumoniae.

Chlamydia pneumoniae (Cpn) could play an important role in the development of atherosclerosis. Cpn interferes with HIF-1alpha regulation in infected host cells during intracellular replication in hypoxia. We obtained carotid artery specimens with low (n=38), high (n=25) levels of stenosis and 10 middle cerebral arteries. Fifty eight percent of the carotids with low levels of stenosis showed evi...
